6-amino-5-(benzylamino)pyrimidine-2,4(1H,3H)-dione, also known as Aminopterin, is a chemical compound used as a medication and as an agricultural herbicide. It belongs to the class of folic acid analogs and works by inhibiting the enzyme dihydrofolate reductase, ultimately preventing the synthesis of DNA and RNA. Aminopterin has been used as a chemotherapy agent for the treatment of leukemia and other malignant tumors, although it is now rarely used due to its toxic side effects. It has also been used as a folate antagonist herbicide to control the growth of weeds, although its use in this context has largely been replaced by safer and more effective compounds. Fragment-based drug design was used to identify Mycobacterium tuberculosis (Mtb) dihydrofolate reductase (DHFR) inhibitors. Screening of ligands against the Mtb DHFR enzyme resulted in the identification of multiple fragment hits with IC50 values in the range of 38–90 μM versus Mtb DHFR and minimum inhibitory concentration (MIC) values in the range of 31.5–125 μg/mL. These fragment scaffolds would be useful for anti-tubercular drug design.
