Doxofylline use and synthesis method
Chemical properties of crystalline, melting point 144-145.5 ℃. Soluble in water, acetone, ethyl acetate, benzene, chloroform, dioxane, hot methanol or hot ethanol, and some insoluble in ether or petroleum ether. Production methodsophylline dissolved in boiling water, sodium hydroxide and 3-chloro-1,2-propanediol, refluxed. Recrystallization from ethanol after evaporation of water gave compound (I) in 95% yield.
The compound (I) is dissolved in water and added with equimolar HIO4 2H2O while stirring, and stirred. The solid was collected by filtration and recrystallized from ethanol to give theophylline acetaldehyde in 70% yield.
Theophylline acetaldehyde, ethylene glycol and p-toluenesulfonic acid reflux dehydration in benzene to give more theophylline, the yield of 70%.
