Pharmacological effects
 
(1) pivastatin had a strong antagonistic effect on HMG-CoA reductase by inhibiting HMG-CoA reductase. The IC50 value was 6.8 nmol/L, which was 24 times stronger than simvastatin and 68 times stronger than fluvastatin.
 
(2) blocking the synthesis of cholesterol pivastatin can effectively inhibit the cholesterol production process in human hepatocytes HepG2, with an IC50 value of 5.8 nmol/L, and its action intensity is 29 times that of simvastatin and 57 times that of avastatin. However, the inhibition of pivastatin on various enzymes in the cholesterol production process after the formation of mevalonate was very weak.
 
(3) increasing LDL receptor density pivastatin can induce the synthesis of LDS receptor mRNA at the ultra-low concentration of 1 microns mol/L, and increase its quantity, leading to the increase of LDL receptor density, thus promoting the removal of LDL, reducing plasma ldl-cholesterol concentration and plasma total triglyceride concentration.
