Attention is called for a superb strategy of peptide synthesis in solution, making use of (per)silylated amino acide fragments.This old methodology may thus present renewed interest if the classical silylation is substituted by silyl cyanides during coupling.Thus, trimethylsilyl cyanide, by its pronounced silylating power affording only gaseous following-(by)products and by its excellent solubilizing properties in organic solvents, overcomes the difficulties and drawbacks that were encountered in previously described strategies that made use of extempore prepared (per)silylated amino acid residues.
