CALCEIN-AM is a hydrophobic, non-fluorescent probe that can permeate the plasma membrane and is hydrolyzed to form Calcein, an extremely fluorescent and negatively charged molecule. It is commonly used as a fluorescent indicator for various applications due to its ability to stain living cells and its use in analyzing cell functions and membrane transport proteins.

Uses

Used in Cell Adhesion Assays:
CALCEIN-AM is used as a cell viability and function indicator for assessing cell adhesion, as it has minimal effects on these parameters.
Used in Cellular Staining:
CALCEIN-AM is used as a cellular stain, allowing for the observation of bright green fluorescence in cytoplasmic Schwann cells and other living cells.
Used in P-glycoprotein and Multidrug Resistance Protein Analysis:
CALCEIN-AM is used as a neutral substrate for P-glycoprotein (Pgp) and multidrug resistance protein (MRP) in flow cytometry studies to analyze the function of these membrane transport proteins.
Used in Liposome Leakage Assays:
CALCEIN-AM is used as a self-quenching fluorescent dye in liposomes, serving as an indicator of lipid vesicle leakage. The leakage rate is calculated based on the fluorescence increment as the entrapped calcein is released from the liposomal compartment.
Used in Calcium Ion Titration and Determination:
CALCEIN-AM is used as a complexometric indicator for titration of calcium ions with EDTA and for fluorometric determination of calcium, with excitation and emission wavelengths of 495/515 nm, respectively.
