Herein, we report a green, expeditious, and practically simple protocol for direct coupling of carboxylate salts and ammonium salts under ACN/H2O conditions at room temperature without the addition of tertiary amine bases. The water-soluble coupling reagent EDC·HCl is a key component in the reaction. The reaction runs smoothly with unsubstituted/substituted ammonium salts and provides a clean product without column chromatography. A general catalytic base-controlled regiodivergent nucleophilic hydrofunctionalization of both terminal and internal β,γ-unsaturated amides has been reported. The atom-economical addition of various S/P-based nucleophiles was also exclusively chemoselective. More than 60 branched or linear hetero-substituted aliphatic amides were synthesized from common starting materials under transition-metal-free conditions. Preliminary mechanistic studies are consistent with our proposed divergent catalytic cycles.
