The invention discloses a preparation method of alogliptin dimer. The preparation method comprises following steps: S1, an intermediate A is prepared, wherein 1-(6-chloro-3-methyl-2,4-dioxo-3,4-dihydro-2H-pyrimidinyl-1-yl methyl)-benzonitrile, (R)-1-N-t-butyloxycarbonyl-3-aminopiperidine, sodium bicarbonate, and isopropanol are uniformly mixed, and are subjected to reflux, and an obtained filtrate is concentrated so as to obtain the intermediate A; S2, an intermediate B is prepared, wherein the intermediate A is added into dichloromethane for dissolving, trifluoroacetic acid is added dropwise, and an obtained mixture is stirred for 1 to 5h, and is subjected to concentration and extraction so as to obtain the intermediate B; and S3, the alogliptin dimer is prepared, wherein the intermediate B, 1-(6-chloro-3-methyl-2,4-dioxo-3,4-dihydro-2H-pyrimidinyl-1-yl methyl)-benzonitrile, potassium carbonate, and N,N-dimethylformamide are uniformly mixed, an obtained mixed material is heated, and is subjected to thermal insulation stirring and filtering, an obtained filter liquor is collected, and is subjected to purifying and recrystallization so as to obtain the alogliptin dimer.
