Description

Fenfluramine hydrochloride, also known as Pondimin, is a crystalline solid that is unique among its group of drugs due to its sedative effects rather than excitatory ones. It is an (±)N-ethyl-α-methyl-3-(trifluoromethyl)phenethylamine hydrochloride, which primarily acts through central serotoninergic mechanisms rather than central noradrenergic ones. Its structure suggests a more apolar or hydrophobic character than amphetamine, indicating a tropism for serotoninergic neurons and an indirect mechanism of action. The increased levels of serotonin lead to greater serotonin receptor activation, which enhances serotoninergic transmission in the hypothalamus, suppressing appetite for carbohydrates. However, it was withdrawn from human use after reports of heart valve damage and pulmonary hypertension.

Uses

Used in Pharmaceutical Industry:
Fenfluramine hydrochloride was used as an appetite suppressant for the treatment of obesity and diabetes. Its action on serotonin receptors, particularly 5-HT1B and 5-HT2C, led to satiety effects and a preference for lower-fat food intake. However, due to its toxicity and withdrawal from the market, alternative treatments are now sought for these conditions.

Methods of preparing a fenfluramine active pharmaceutical ingredient are provided. Aspects of the method include (a) hydrolyzing a 2-(3-(trifluoromethyl)phenyl)acetonitrile composition to produce a 2-(3-(trifluoromethyl)phenyl)acetic acid composition; (b) reacting the 2-(3-(trifluoromethyl)phenyl)acetic acid composition with acetic anhydride and a catalyst to produce a 1-(3-(trifluoromethyl)phenyl)propan-2-one composition; and (c) reductively aminating the 1-(3-(trifluoromethyl)phenyl)propan-2-one composition with ethylamine using a borohydride reducing agent to produce a fenfluramine composition. Also provided are compositions and pharmaceutical ingredients prepared according to the subject methods including a pharmaceutically acceptable salt of fenfluramine and having less than 0.2% by weight in total of trifluoromethyl regioisomers.
