The first total synthesis of the methyl ester of 20-oxo-LTB4 26 is described. The key synthon 6 is an advanced new intermediate which has been used in the synthesis of LTB4 1, 20-oxo-LTB4 methyl ester 26, and 20-hydroxy-LTB4 2. The synthetic 26 has been used to study the cytosolic aldehyde dehydrogenase-catalyzed oxidation of LTB4 to its ω-carboxy metabolite.
