Enzymic synthesis of glaucine in cell free system from Litsea glutinosa

The bioconversion of 1-benzyltetrahydroisoquinolines such as norcoclaurine (1), coclaurine (2), N-methylcoclaurine (3), 6-methoxynorlaudanosoline (4), 6-methoxylaudanosoline (5) and reticuline (6) and aporphine alkaloids isoboldine (11), thaliporphine (12) and N-methyllaurotetanine (13) into glaucine (14) in the cell free system from Litsea glutinosa (Lour) C.B.Rob. var. glabraria Hook f. (Lauraceae) has been studied.Stereospecificity of enzymes has been demonstrated by parallel incubation of (S)-(+)-, and (R)-(-)-reticulines.The incubation experiments reveal thatglaucine is synthesized by trihydroxylated base.N-Methylation of 2 is preferred over aryl hydroxylation to form 6 which by oxidative coupling forms 11 and methylation via 12 gives glaucine (14).
