Description

ALPHA,ALPHA-TREHALOSE GLUCOHYDROLASE, also known as Trehalase, is a glycosidase enzyme that belongs to the glycoside hydrolase 37 (GH37) family of the carbohydrate-active enzyme (CAZy) classification. It is found in many forms and occurs in all tissues, playing a crucial role in the breakdown of trehalose, a disaccharide sugar, into its constituent monosaccharides, glucose.

Uses

Used in Plant Pathology Research:
ALPHA,ALPHA-TREHALOSE GLUCOHYDROLASE is used as a research tool for assessing changes in carbohydrate metabolism in Plasmopora viticola-infected grapevine leaves. This helps in understanding the impact of infections on plant metabolism and developing potential treatments.
Used in Cellular Biology Studies:
In cellular biology, ALPHA,ALPHA-TREHALOSE GLUCOHYDROLASE is used as an investigative tool to study growth arrest mechanisms regulated by trehalose-6-phosphate. This aids in understanding the role of trehalose-6-phosphate in controlling cell growth and its potential implications in various biological processes.
Used in Microbiology:
ALPHA,ALPHA-TREHALOSE GLUCOHYDROLASE is utilized as a quantification method for determining the intracellular trehalose levels in Lactobacillus casei cells. This application is vital for microbiological research, allowing scientists to study the metabolic activities and stress responses of these microorganisms.

Biochem/physiol Actions

Trehalase catalyzes the hydrolysis of trehalose to produce two glucose units. It mediates flight metabolism, molting induced chitin production and tolerance towards cold temperature.

Purification Methods

This trehalase is purified by solubilising in Triton X-100 and sodium deoxycholate, and submitting to gel filtration, ion-exchange chromatography, conA-Sepharose chromatography, phenyl-Sepharose CL-4B hydrophobic interaction chromatography, Tris-Sepharose 6B affinity and hydrolyapatite chromatography. Activity is increased 3000-fold. [Yoneyama Arch Biochem Biophys 255 168 1987.] T4-RNA ligase (from bacteriophage-infected E.coli ) Mr 43,500, [EC 6.5.1.3 for RNA lyase]. This ligase is purified by differential centrifugation and separation on a Sephadex A-25 column, then through hydroxylapatite and DEAE-glycerol using Aff-Gel Blue to remove DNAase activity. (Greater than 90% of the protein in the enzyme preparation migrated as a single band on gradient polyacrylamide gels containing SDS during electrophoresis.) [McCoy et al. Biochim Biophys Acta 562 149 1979.]

Check Digit Verification of cas no

The CAS Registry Mumber 9025-52-9 includes 7 digits separated into 3 groups by hyphens. The first part of the number, starting from the left, has 4 digits, 9,0,2 and 5 respectively; the second part has 2 digits, 5 and 2 respectively.
Calculate Digit Verification of CAS Registry Number 9025-52:
(6*9)+(5*0)+(4*2)+(3*5)+(2*5)+(1*2)=89
89 % 10 = 9
So 9025-52-9 is a valid CAS Registry Number.
