General Description

1-[3-(4-chlorophenyl)-5-isoxazolyl]-1-ethanone, also known as Etazene, is a synthetic opioid analgesic drug with potent and fast-acting pain-relieving properties. It is structurally similar to fentanyl and is considered to be one of the strongest opioid agonists available. Etazene works by binding to the body's opioid receptors, which are responsible for modulating pain and pleasure sensations. Due to its high potency, Etazene has a high potential for abuse and addiction, and has been associated with several overdose deaths. As a result, it is classified as a controlled substance and is not approved for medical use in most countries.

Hydroxy (tosyloxy) iodobenzene (HTIB), a hypervalent iodine reagent, has been extensively used for oxidative transformations. We have developed a one-pot synthesis wherein aldoximes when reacted with alkynes in the presence of HTIB result in the direct formation of isoxazoles. This simple and straightforward reaction allows for ease of purification while leading to the formation of high purity 3,5-disubstituted isoxazoles in moderate yields.
