Thioredoxin reductase (TrxR) enzymes are critical in regulating redox homeostasis in cells. We report the first two-photon fluorescent probe of mammalian TrxR (TP-TRFS). TP-TRFS retains high specificity in recognizing TrxR. More importantly, the two-photon absorbing character of TP-TRFS enables it to be used in vivo. With the aid of TP-TRFS, a remarkable decline of the TrxR function was observed in the brain of a mouse model of stroke for the first time, providing a mechanistic link of TrxR dysfunction with stroke. The invention discloses a PRODAN fluorescent dye with red fluorescence and large Stokes shift, and a synthesis method and an application thereof. The dye has a structure defined in the specification, wherein R1 is one of C1-20 alkyl, substituted alkyl, cycloalkyl, substituted cycloalkyl, aryl, substituted aryl, heteroaryl, substituted heteroaryl, hydroxyl, cyano or alkoxy; R2, R3, R4 and R5 are respectively one or two or more of H, C1-20 alkyl, substituted alkyl, cycloalkyl, substituted cycloalkyl, aryl, substituted aryl, (CH2)nOH, (CH2)nCOOM, (CH2)nSO3M, heteroaryl or substituted heteroaryl; n is an integer of 0-12; M is one or two or more of H, K, Na, Li, NH4, NH3R6, NH2(R6)2, NH(R6)3 or N(R6)4; R6 is one or two or more of H and C1-20 alkyl. The obtained dye has relatively high light stability, has the fluorescence emission wavelength at about 582 nm, also has relatively large Stokes shift and relatively high fluorescence quantum yield, and can be applied in luminescent materials, biological fluorescent probes, biological fluorescent imaging and other fields.
