Tenovin-1 is a compound that belongs to the class of Tenovins, and inhibits SIRT1 and SIRT2. SIRT1 and SIRT2 are protein-deacetylases and are members of the NAD+-dependent class III histone deacetylases. Tenovin-1 has been shown to activate and elevate acetylated p53 levels, induce apoptosis in vitro.
in vitro: Tenovin-1 elevates the amount of p53 protein within 2 hr of treatment. This compound also increases the levels of the p53-downstream target p21CIP/WAF1 protein and mRNA, confirming that tenovin-1 can induce expression from an endogenous p53-dependent promoter [1]. Tenovin-6 inhibits the growth of S. cerevisiae cultures with an IC50 of 30 μM and is more toxic to yeast than the less water-soluble tenovin-1.
in vivo: Initial in vivo experiments indicated that tenovin-1 impairs the growth of BL2-derived tumor xenografts [1].
