About:
Methylprednisolone is a glucocorticoid drug, used for emergency treatment of critical diseases, endocrine disorders, rheumatic diseases, collagen diseases, skin diseases, allergic reactions, eye diseases, gastrointestinal diseases, blood diseases, leukemia, shock, brain edema, multiple neuritis, myelitis and to prevent vomiting caused by cancer chemotherapy.
In vivo studies:
Methylprednisolone reduces RGC survival in rats with electrophysiologically diagnosed optic neuritis. Methylprednisolone reduces RGC survival by a non-genomic, calcium-dependent mechanism. Methylprednisolone induced enhancement of RGC denaturation depends on calcium inflow through volt-gated calcium channels. In beak and tail stumps, treatment with Methylprednisolone resulted in a significant reduction in the number of ED1-positive cells.
Methylprednisolone resulted in a significant reduction in tissue loss after treatment of 2,4 and 8 after one week of injury. After 1 and 2 weeks of transitional injury, Methylprednisolone resulted in long-term reduction of ED1-positive cells and loss of spinal cord tissue, reduced vestibular fiber dieback, and instantaneous germination of vestibular fibers adjacent to the lesion. In a rat model of spinal cord injury, Methylprednisolone (30mg/kg) was effective in improving functional outcomes, inhibiting TNF-α expression and NF-kB activation. Methylprednisolone inhibits the function of NF-kB, possibly induced by IKB, by trapping NF-kB in inactive cytoplasmic complexes.
