Irinotecan hydrochloride is a semi synthetic water-soluble camptothecin derivative with light yellow powder or pale yellow crystalline powder at room temperature without odor. Slightly soluble in water, ethanol or chloroform, insoluble in acetone. CPT can specifically combine with topoisomerase I, which induced reversible single strand breaks, so that the double strand of DNA helicase; irinotecan and its active metabolite SN-38 can bind topoisomerase I-DNA complexes, and then connected to prevent fracture bond, resulting in cell toxicity, the cytotoxicity is time dependent, and specific to the S phase, the toxicity is also stronger than irinotecan. Suitable for the treatment of patients with advanced colorectal cancer, and the combined treatment of 5- fluorouracil and folinic acid had not received chemotherapy for patients with advanced colorectal cancer, as monotherapy, treated by 5- containing fluorouracil chemotherapy treatment failure patients. Irinotecan has broad spectrum and strong antitumor activity both in vivo and in vitro. It is important that irinotecan and its metabolites are still effective in the expression of multidrug resistant tumors.
