Gramicidin is a heterogeneous mixture of three antibiotic compounds, gramicidins A, B and C, making up 80%, 6%, and 14%, respectively,all of which are obtained from the soil bacterial speciesBacillus brevis and called collectively gramicidin D. Gramicidin D contains linear pentadecapeptides, that is chains made up of 15 amino acids. This is in contrast to gramicidin S, which is a cyclic peptide chain.Gramicidin from Bacillus aneurinolyticus (Bacillus brevis) is a linear polypeptide antibiotic with a mixture of gramicidin A, B, C, and D. Gramicidin D is a channel-forming ionophore that flip-flops slowly across the membrane and is a known Pgp substrate and surprisingly was found to inhibit Pgp ATPase activity. This inhibition was reversed by other Pgp substrates suggesting a common drug binding site among MDR substrate-type drugs and chemosensitizers.

Function                                          

Gramicidin is active against Gram-positive bacteria, except for the Gram-positive bacilli, and against select Gram-negative organisms, such as Neisseria bacteria. Its therapeutic use is limited to topical application, as it induceshemolysis in lower concentrations than bacteria cell death, so it cannot be administered internally. Since the exterior epidermis is composed of dead cells, applying it to the surface of the skin will not cause harm.

It is used primarily as a topical antibiotic and is one of the three constituents of consumer antibioticpolysporinophthalmic solution.

Application                                            

It is a lipophilic substance, the two short bacitracin molecules are dimer to the left helix space configuration, constitute a cross-lipid membrane membrane channel to combine and carry a specific cation from the side of the membrane into the channel and then spread to The other side of the membrane, so there is the name of ion antibiotics, so that the cell metabolism can not be normal and inhibited or even death.
