As a high molecular polymer, serum separation gel has the advantages of insoluble in water, oxidation resistance, high temperature resistance, low temperature resistance and good air tightness. It has been widely used in blood collection tubes.
Because there are a lot of hydrogen bonds in the separation structure, when the separating glue and the solidified blood are centrifuged in the same tube, due to the centrifugal force, the hydrogen bonded network structure is destroyed into a chain structure and becomes a low viscosity fluid. When the centrifugal force is removed, the chain structure in the separation glue is reconstituted into a network structure and restored to a high viscosity gel state. It is normal to form an isolation layer between serum and red blood cells due to density. However, if the isolation layer is not formed, but the separation glue floats Separation gel and red blood cells mixed together can not be separated What causes this
Separation glue floating phenomenon
1. The position of serum separation gel is caused by plasma or serum density, not viscosity. Any significant increase in plasma or serum density will cause the separation gel to float.
2. Monoclonal immunoglobulin disease (MGP) produces a large amount of immunoglobulin, which causes the change of serum protein, and the increase of protein will also cause the increase of serum density. Therefore, when the protein increases to a certain level, the serum density of blood samples collected by blood collection vessels with separation gel can be higher than that of separation gel, and the separation gel will float at this time.
3. Patients with contrast agent and blood samples collected through dialysis tube can have separation gel floating phenomenon. The reason is that the increase of serum density caused by contrast agent and dialysate is higher than that of separation gel, resulting in colloidal floating.
4. The use of iodized contrast medium for coronary angiography and intervention will cause the floating phenomenon of separation gel, and the phenomenon that serum and red blood cells can not be separated by mixing, indicating that contrast medium can not only increase serum density, but also cause poor blood clot contraction.
Separation glue and red blood cells cannot be separated
1. It is difficult to separate the mixed red blood cells and serum. When a certain proportion is reached, the mixed density of red blood cells and serum is close to the density of separation gel, which will lead to poor separation of red blood cell gel.
2. Low speed centrifugation can lead to poor separation of separation gel and red blood cells, while higher speed centrifugation can correct some poor separation phenomena. When the separation gel and red blood cells are still poorly separated after higher speed centrifugation, the possibility of MGP in the patient should be considered. On the contrary, mpg will also cause ineffective separation of serum and red blood cells, resulting in poor separation of separation gel and red blood cells.
