Details:

Sinomenine is an alkaloid extracted from the dried cane of Caulis Sinomenii.
(1) Analgesic effect: Sinomenine has proved to have a certain analgesic effect in the experiments of mouse electrical stimulation method, hot plate method and rabbit photothermal stimulation method. The site of its analgesic effect is in the central nervous system, the dose required for analgesia is about 10 times that of morphine, the duration is short, and continuous application can also produce tolerance. Sedative effect: Sinomenine significantly reduced the spontaneous and passive activities of mice, but had no significant effect on barbiturate sleep time. Sinomenine has an antitussive effect, and the antitussive potency is similar to that of codeine; it has a slight emetic effect.
(2) Anti-inflammatory effect: Sinomenine has a significant regressive effect on formaldehyde-induced and egg white-induced "arthritis" in rats, but has no such effect after removal of the adrenal gland or pituitary gland. Therefore, the principle of anti-inflammatory effect may be caused by the influence of the hypothalamus on the pituitary-adrenal system, and has nothing to do with the release of histamine. It has a preventive effect on active anaphylactic shock in guinea pigs, but it is relatively poor in dogs.
(3) Sinomenine has antihypertensive effect. Oral or intravenous injection has antihypertensive effect on normal rats, dogs, anesthetized cats and dogs with chronic renal hypertension; It is related to its anti-adrenergic effect and inhibition of barotropic reflex mechanisms. Sinomenine also has a certain antagonistic effect on several different types of arrhythmia models.
(4) Sinomenine has obvious antitussive effect, and its potency on rats and cats is about the same as that of codeine. It has a powerful histamine release effect on the body. Experiments also preliminarily show that Sinomenine has obvious stimulating effect on cellular immunity; there are also reports that Sinomenine has inhibitory effect on non-specific and specific immunity of the body.
(5) Effects on gastrointestinal activity: Oral administration of sinomenine to dogs and monkeys often has mild gastrointestinal adverse reactions; it has inhibitory effects on isolated rabbit intestines and guinea pig intestines, and can resist pilocarpine, histamine, and acetylcholine. and the spasmodic effect of barium chloride. But for the dog and rabbit intestines, intravenous injection of sinomenine can cause temporary excitement of the small intestine. Injection of sinomenine can increase gastric secretion and acidity, but the activity of pepsin has no significant change. The excitatory effect on the gastrointestinal tract is mainly related to the release of histamine. The acidic extract of fenugreek can excite isolated rabbit intestines.
