Entecavir is a new generation of oral guanosine analogs for the treatment of hepatitis B virus infection. It is mainly used for the treatment of chronic hepatitis B with active viral replication and persistently elevated serum transaminases, or active liver histology in adults. Hepatitis is currently the fastest and strongest nucleoside analog for virus reduction and the lowest mutation probability. The data show that: different chronic hepatitis B patients, including nucleoside-naïve, nucleoside-experienced and liver cirrhosis patients, can quickly control the disease by using entecavir tablets, and easily reach the realistic endpoint of treatment, that is, hepatitis B virus is undetectable; After treatment, a considerable number of patients can reach the satisfactory end point of treatment, that is, e-antigen seroconversion, and some patients can even reach the ideal end point of treatment, that is, surface antigen negative conversion.

This product is a guanosine analog, which has inhibitory effect on hepatitis B virus (HBV) polymerase. It can be phosphorylated into an active triphosphate with an intracellular half-life of 15 hours. By competing with deoxyguanosine triphosphate, the natural substrate of HBV polymerase, entecavir triphosphate inhibits all three activities of viral polymerase (reverse transcriptase): (1) HBV polymerase initiation; (2) pregenomic mRNA The formation of reverse transcription negative strand; (3) the synthesis of HBVDNA positive strand. The inhibition constant (Ki) of entecavir triphosphate to HBV DNA polymerase was 0.0012 μM. Entecavir triphosphate has weak inhibitory effect on α, β, δ DNA polymerase and mitochondrial γ DNA polymerase in cells, and the Ki value is 18 to 160 μM.
