Bile acid is a general term for a class of cholanoic acids present in bile. It is a white powder, odorless and bitter in taste. Its alkali metal salts are easily soluble in water and alcohol. Natural bile acids are usually combined with glycine or taurine by peptide bonds and combined with sodium and potassium ions to form bile salts in bile. Bile acids in normal human bile can be divided into two categories according to their structure: one is free bile acids, including cholic acid, deoxycholic acid, chenodeoxycholic acid and a small amount of lithocholic acid. Acid (lithochalicacid); another type is the product of the above-mentioned free bile acid combined with glycine or taurine, called conjugated bile acid. It mainly includes glycocholic acid, glycochenodeoxycholic acid, taurocholic acid and taurochenodeoxycholic acid. Generally, the bound bile acid is more water-soluble than the free type, and this combination makes the bile acid salt more stable, and it is not easy to precipitate out in the presence of acid or Ca2+. Bile acids can be divided into primary bile acids and secondary bile acids according to their different sources in the body. In liver cells, bile acids directly synthesized from cholesterol are called primary bile acids, including cholic acid and chenodeoxycholic acid. Both cholic acid and chenodeoxycholic acid are cholanoic acid derivatives containing 24 carbon atoms. The difference in structure between the two is only the number of hydroxyl groups, cholic acid contains 3 hydroxyl groups (3α, 7α, 12α), while chenodeoxycholic acid contains 2 hydroxyl groups (3α, 7α). Primary bile acids are subjected to bacterial action in the intestinal tract, and bile acids generated by 7α dehydroxylation are called secondary bile acids, including deoxycholic acid and lithocholic acid. The three-dimensional configuration of bile acids has two sides, hydrophilic and hydrophobic, so that bile acids show strong interfacial activity. It can reduce the surface tension between the lipid and water phases, and promote the formation of mixed micelles of lipids, which plays an important role in the digestion and absorption of lipids and the maintenance of the dissolution of cholesterol in bile. Thin-layer chromatography methods for cholic acid, dehydrocholic acid, chenodeoxycholic acid, ursodeoxycholic acid, isodeoxycholic acid and lithocholic acid: chromatography plate, silica gel G plate; sample, prepared with methanol into a 1mg/ml solution; developing agent, isooctane-diethyl ether-glacial acetic acid-n-butanol-water (10:5:5:3:1); developer: 30% sulfuric acid solution, dry at 105°C after spraying Bake for 10 min and observe fluorescence under 365 nm UV light.
