Dacarbazine is a new type of purine precursor antineoplastic drug, which can interfere with the biosynthesis of purines and also act as an alkylating agent. It has inhibitory effect on mouse sarcoma-180, lung cancer-755, melanoma B16 and leukemia L-1210. It acts on the G2 phase of the cell cycle, mainly affecting RNA and protein synthesis, followed by DNA. Oral absorption is incomplete, and individual differences are large. After 1 intravenous injection, it reached a peak value within 30 minutes and disappeared. 30% of the dose was excreted in the urine in 6h. Cannot pass the blood-cerebrospinal fluid barrier. The citrate salt of dacarbazine, white crystalline powder, is mainly used clinically. The aqueous solution is highly acidic, with a pH of 1.9 to 2.3. Mainly used for malignant melanoma, better than hydroxyurea, such as combined with vinblastine, carmustine can improve the efficacy. Also used for lung squamous cell carcinoma and undifferentiated carcinoma, leiomyosarcoma, fibrosarcoma and so on. Ineffective for gastrointestinal tumors.

For disseminated malignant melanoma, the curative effect is better than other anticancer drugs. Single-agent therapy is most commonly used, but it can also be combined with BCG, transfer factor, and milfalan for immunochemical combination therapy. With (ABVD) and (MOPP), for some patients with advanced Hodgkin's disease with poor prognosis, alternate two kinds of combined chemotherapy can achieve long-term complete remission and even cure. It has a certain curative effect on various sarcomas, brain tumors, lung squamous cell carcinoma and small cell carcinoma, as well as childhood neuroblastoma.
