Target point
AGE [1]
In vitro studies have shown that Alagebrium chloride is an inhibitor of advanced glycation end products (AGE). The proliferation of endothelial cells (EC) increased in all groups receiving Alagebrium (ALT-711), especially when the matrix was inoculated from AAo of obese (ZO) and diabetes (ZD) rats [2].
In vivo research using Alagebrium treatment does not affect blood pressure. In RAGE apoE double KO mice with diabetes, treatment with Alagebrium was associated with a modest reduction in renal mass and reduced ultrafiltration compared with untreated mice. Treatment with Alagebrium in RAGE apoE double KO mice with diabetes is related to further reduction of glomerular collagen IV level, close to the level observed in control mice [1]. In the absence or presence of Alagebrium (ALT-711), Zucker thin (ZL), obese (ZO) and diabetes (ZD) groups had similar body weight, heart rate (HR) and mean blood pressure (BP). Alagebrium increases blood flow (BF) in ZO rats but reduces distal vascular resistance in ZD rats. In all groups treated with Alagebrium, changes in neointimal hyperplasia (NH) and endometrial thickness with local radius were observed. TGF was also found in AAo of ZL rats treated with Alagebrium- β Significant increase in expression [2].
Cell experiment Human aortic endothelial cells (HAECs) were seeded with acellular matrix derived from abdominal aorta (AAo) of Zucker lean (ZL), obese (ZO) and diabetes (ZD) rats, with or without Alagebrium (ALT-711) (20 μ g) In the presence of 1 × Antibiotics -/mL of Dulbecco's PBS for antifungal agents. Conduct the experiment when the cells reach 80 to 90% convergence. The flow chamber is sealed to the HAEC monolayer through a vacuum network. The flow is driven by a Masterflex L/S peristaltic pump, and the humidification chamber is heated to 37 ° C for 4 hours. Leibovitz-15 medium supplemented with 10% FBS, endothelial BulletKit, and 1 × Antibiotic antifungal solution, used as a mobile culture medium to maintain pH in the absence of CO2 [2].
Animal experiments randomized RAGE apoE mice to be treated with Alagebrium (1mg/kg/day via tube feeding) or untreated (n=20/group). After 20 weeks of diabetes, mice were placed in individual metabolic cages for 24 hours and urine was collected. Record weight and fluid and food intake. According to the reagent kit protocol, the mouse white
