(2S) -n-chloroacetyl - 2-cyano-tetrahydropyrrole is the intermediate for the synthesis of Vigliptin, mainly used for the synthesis of vigliptin, such as L-proline amide as raw material, with THF as solvent, K2CO3 as acid binding agent, chlorine acetylation reaction, to obtain (S)-1-(2-chloroacetyl) -2-pyridine-formamide Chemicalbook, Then, through dehydration cyanidation reaction with TFAA, the amide group was converted to nitrile, and the intermediate (2S) -n-chloroacetyl - 2-cyano-tetrahydropyrrole was obtained. Finally, DCM was used as the solvent, K2CO3 was used as the acid binding agent, and the condensation reaction was carried out with 3-amino-1-amantadanol, and then the column chromatography was used to refine the Vegliptin
