alogliptin benzoate (also known as alogliptin) is a serine protease dipeptidyl peptidase Ⅳ(DPP-Ⅳ) inhibitor developed by Takeda company in Japan. This product can maintain the levels of glucagon-like peptide 1(GLP-1) and glucose-dependent insulin stimulating polypeptide (GIP) in the body, promote the secretion of insulin, and thus play a hypoglycemic role. Results of a randomized, placebo-controlled clinical trial showed that alogliptin monotherapy significantly improved glycemic control in patients with type 2 diabetes compared with placebo, did not increase the incidence of hypoglycemia, and was well tolerated. Alogliptin and its drug compositions are specific and effective oral DPP-Ⅳ inhibitors after sitagliptin and Vigagliptin, which provide a new and important treatment option for type 2 diabetes patients and have a wide application prospect. Alogliptin benzoate was officially launched in China in early 2014 and is the first type 2 diabetes treatment drug approved in China with proven cardiovascular safety data, which provides more and better options for the treatment of type 2 diabetes.

Synthesis method

(1) Using 3-methyl-6-chlorouracil as starting material, alkylation reaction with 2-bromomethylbenzylnitrile under alkaline conditions, and then substitution reaction with (R) -3-aminopiperidine dihydrochloric acid to obtain (R)-2-[(6-(3-aminopiperidine-1-yl) -3-methyl-2, 4-dioxo-3, Alogliptin benzoate was prepared from 4-dihydropyrimidine-1 (2H) -methyl] benzylnitrile (A01), which was then salt with benzoic acid. The total absorption rate of this method is 56.1%, the raw materials are easy to obtain, the reaction conditions are mild, the purity is good, the kinds of reagents are few and so on, and it is more suitable for industrial production.

(2) 6-chlorouracil was used as starting material to alkalize with 2-bromomethylbenzylnitrile under the action of sodium hydride and lithium bromide, and then reacted with iodomethane under the action of sodium hydride to substitute with (R) -3-aminopiperidine dihydrochloride under alkaline conditions to obtain (A01), and (A01) was salted with benzoic acid to obtain alogliptin benzoate. The total absorption rate of this method can reach 27.2%, but sodium hydride and lithium bromide are used in large quantities, and the reaction requires high anhydrous and anaerobic conditions. On the other hand, the toxicity of iodomethane is not conducive to environmental protection and is not suitable for industrial production.
