overview | astragalus is the dried root of legume astragalus mongholicus (Astragalusmembranaceus.) Bge.var.mongholicus(Bge.)Hsiao) or membranous astragalus (Fisch.) Bge.). Astragalus is a perennial herb, which is resistant to cold, drought, heat and waterlogging. It mostly grows in arid sunny hillside grassland or sunny forest edge shrubs. Cultivation should be in a cold and dry climate, with sandy soil with good drainage and deep soil layer. Mongolian Astragalus membranaceus is distributed in some provinces and regions of Northeast and North China; Astragalus membranaceus is distributed in some provinces and regions of North, Northwest, Northeast and Southwest China. Astragalus is a commonly used Chinese medicine. It is warm in nature and sweet in taste. It has the functions of replenishing qi and strengthening the surface, diuresis and supporting poison, discharging pus, and gathering sores and muscles. It is used for qi deficiency and fatigue, eating less and loose stools, sinking of medium qi, and prolapse of anus for a long time., Blood in the stool, bleeding and leakage, superficial self-sweating, qi deficiency and edema, long-term collapse and failure, chronic nephritis, proteinuria, diabetes and other diseases. Modern pharmacological studies have shown that Astragalus has the effects of enhancing the body's immune function, strengthening the heart and lowering blood pressure, lowering blood sugar, diuresis, anti-aging, and anti-fatigue. The main active ingredients in Astragalus membranaceus are astragalus polysaccharide (astragaluspolysaccharides), astragaloside (astragalussaponins) and astragalus isoflavone (isoflavones). At present, astragaloside IV (astragaloside IV) in astragaloside is mainly used as the standard to evaluate the quality of astragalus membranaceus. |
Use | Astragaloside A is an isomer of Astragaloside IV. Astragaloside IV has protective effects on cardiovascular, immune, digestive and nervous system. |
pharmacological action | 1. astragaloside IV can inhibit oxidative stress, reduce the level of reactive oxygen species (ROS) in cells, enhance antioxidant defense system, increase anti-apoptotic and anti-inflammatory pathways, and regulate the differentiation and penetration of T cells in the central nervous system. This suggests that astragaloside IV may be clinically used for the treatment and prevention of MS. In short, astragaloside IV can play a certain role in many diseases by regulating the immune function of the body. 2. Astragaloside IV has protective effects on brain injury, lung injury, kidney injury and heart injury caused by ischemia reperfusion. 3. Astragaloside IV has a protective effect on myocardial injury caused by ischemia and hypoxia, and it has a certain effect on myocardial hypertrophy, myocarditis, and myocardial fibrosis. 4. Astragaloside IV has an effect on pneumonia, airway inflammation, asthma, acute pancreatitis, etc. through anti-inflammatory activity. 5. Astragaloside IV plays an anti-CVB3 virus effect by up-regulating the expression of IFN-γ at mRNA level. 6. Astragaloside IV inhibits the activities of hepatic glycogen phosphorylase (GP) and glucose -6-phosphatase (G6Pase), induces TNF-α secretion in 3T3-L1 adipocytes, and improves insulin tolerance. 7. Astragaloside IV has certain inhibitory effects on lung cancer, liver cancer, breast cancer and gastric cancer in vitro. |
toxicity | to study the effect of astragaloside iv on embryo and fetal development of SD rats and New Zealand rabbits. The results showed that when the dosage of was more than 1mg kg-1, astragaloside IV showed toxicity to the mother. When the dose is greater than 0.5mg kg-1, it shows toxicity to the fetus. Therefore, it is recommended that pregnant women use astragaloside IV carefully. Further studies have shown that astragaloside IV can significantly delay hair development, eye opening time and Parry reflex in rats. However, astragaloside IV had no significant effect on memory and learning ability. |
preparation | enzymatic hydrolysis of astragaloside glycosides to prepare astragaloside IV: a. Aspergillus nigrans (Aspegillusniger) is stirred and ventilated for 50-100 hours at 28-30 ℃ in a medium containing 5% malt water extract and 1% enzyme-producing inducer-astragalus water extract, centrifugal sterilization to obtain the supernatant (mixed solution containing enzyme), in the supernatant with 60-75% saturation of ammonium sulfate precipitation enzyme protein, protein collection, dissolved in 1/10 of the fermentation broth volume of commonly used acetic acid buffer (0.02M,pH5.0), or phosphate buffer, Tris buffer, dialysis to remove ammonium sulfate, centrifugal slag, is the enzyme solution; B. mix 50 ml of the above enzyme solution, 3g of astragaloside and 100 ml of acetic acid buffer (0.02M,pH5.0) evenly, react at a temperature of 20~55 ℃ for 6~24 hours, then add 1/3 volume of n-butanol to extract saponin for three times, and evaporate to dry under reduced pressure to obtain crude astragaloside. 0.5~0.8 grams of astragaloside were obtained by silica gel column separation method (document 3). the conversion rate of astragaloside reached 60 ~ 90%, which was 6 times higher than that of astragaloside directly extracted from the original 3 grams of astragaloside. The above enzyme solution was separated and purified by DEAE-Cellulose ion exchange resin column method and BioRed protein preparation chromatograph, and the molecular weight of the enzyme was determined by SDS electrophoresis, and the molecular weight of the enzyme was 50000. |
