Octreotide acetate is a long-acting synthetic analog of natural somatostatin and is a more potent inhibitor of growth hormone, glucagon, and insulin than somatostatin.

In vitro activity: The octreotide-treated group showed a significant reduction in tumor volume compared with the saline group. Octreotide-PPSG (1.4 mg/kg, intraperitoneally) showed a stronger antitumor effect than octreotide solution (100 μg/kg, intraperitoneally). Octreotide treatment had a significant inhibitory effect on the expression levels of SSTR2 and SSTR5 in rats bearing primary HCC compared with the saline group. Octreotide-PPSG appeared to suppress the expression of SSTR2 and SSTR5 more than the octreotide-solution-treated group. The tested dose of octreotide acetate significantly reduced serum gastrin levels to approximately one-third of baseline within 2 hours, and the effect lasted for approximately 6 hours. On day 21, treatment with a sustained-release formulation of octreotide acetate (5 mg intramuscularly, q 4 weeks) was initiated.
