Introduction	Sesamin is a natural ingredient contained in sesame seeds, which contain about 0.3-0.5% sesamin. Sesamin has excellent antioxidant effect, can be anti-aging, regulate blood lipids, protect the liver, lower blood pressure, anti-cancer, regulate immunity, make the hair black and bright and many other physiological effects. In human experiments, it is proved that sesamin can be transported to the liver and metabolized into antioxidant substances in the liver, and the antioxidant effect is stronger than that of vitamin E.
preparation	take 1L of sesame oil foot (965.8g), add 4L of anhydrous ethanol, the mixture was stirred at 30 ° C. The crude sesamin was dissolved in anhydrous ethanol, and the ratio of crude sesamin to anhydrous ethanol was 1:30(kg/L), time to maintain 3 hours, in this condition after 2 times crystallization, vacuum drying to obtain sesamin product 2.16g, sesamin extraction rate 89.1%, and the purity of the obtained sesamin product was 92.1%.
Content determination	a method for determining the content of sesamin in sesame seeds, comprising the following steps:(1) preparation of sesamin standard solution: weigh 10mg of sesamin standard, add anhydrous methanol to dissolve and dilute to 50mL, then filter with 0.45 μm filter membrane to obtain 0.2mg/ mL of sesamin standard solution;(2) draw the standard curve of sesamin: take 20, 10, 5, 4, 2, 0.8 of the sesamin standard solution obtained in step (1), and add anhydrous methanol 0, 10, 15, 16, 18, 19.2mL, shake to prepare the methanol solution of sesamin standard with gradient concentration of 200, 100, 50, 40, 20, 8ug/mL, the above solutions were analyzed by liquid chromatography, and the standard curve of sesamin was drawn according to the determination results (the liquid chromatogram of sesamin standard solution is shown in Figure 1), the linear relationship equation between the content of sesamin and the peak area is obtained: A = 587.525675x-4.531582, where A is the peak area and x is the content of sesamin;(3) prepare the sample solution to be tested: using Yuzhi No. 11 sesame seeds as the sample to be tested, the sesame seeds to be tested were crushed by a universal pulverizer for 30s, then sieved through a sieve of 0.35 μm, and 0.2g of the obtained sesame powder sample was taken, add 100% ethanol 5ml;(4) ultrasonic treatment and filtration: the mixed solution obtained in step (3) in 30 ~ Under the condition of 60 ℃, ultrasonic treatment was carried out for 20~50min, and then the solution after ultrasonic treatment was filtered with 0.45 μm filter membrane, the model is KQ3200DE CNC ultrasonic cleaner, ultrasonic processing is carried out under the ultrasonic power of 70~100W. (5) detection: Take 9 μl of the filtrate obtained in step (4), use liquid chromatography to detect the filtrate, according to the standard curve of sesamin, the content of sesamin in sesame seeds was calculated by the linear equation.
