In vitro study |
In intact cells, PPP potently inhibited IGF-1 of Akt (Ser 473) and Erk1/2 phosphorylation stimulated by IGF-1R. In IGF-1R positive tumor cells in culture, picropodophylin specifically inhibited cell growth and induced apoptosis. Picropodopylin synergistically sensitized HMCL, primary human MM and mouse 5T33MM cells to ABT-737 and ABT-199 by further reducing cell viability and enhancing apoptosis. Picropodopylin and sorafenib synergistically inhibit the proliferation and activity of hepatocellular carcinoma. |
In vivo study |
In SCID mice xenografted with human ES-1,BE, and PC3, Picropodophyllin (20 mg/kg/12 h, I. p.) caused complete tumor regression. In the 5t33mm mouse model, Picropodophyllin also showed potent anti-tumor activity and significantly increased survival. |
