LGD-3303 is a non-steroidal, non-aromatic androgen receptor ligand that binds the androgen receptor with high affinity (Ki=0.9nM) in a radiolabeled competitive binding assay. Compared with the salocorticoid, glucocorticoid and progesterone receptors, its affinity is greatly reduced, with Ki= 1261,581 and 136nM, respectively. LGD-3303 effectively activates transcription through androgen receptors (EC50=3.6nM) with 134% efficacy relative to the steroid androgen dihydrotestosterone (DHT) 1.

 

R&d and application

LGD-3303 has been studied as a potential drug for the treatment of osteoporosis and has shown the ability to enhance the effects of bisphosphonate drugs in animal studies. It acts as a selective androgen receptor antagonist with good oral bioavailability and is able to inhibit muscle weight loss in vivo and increase muscle weight beyond eugonadal levels at higher dosages 1.

 

pharmacokinetics

In one study, LGD-3303 completely suppressed muscle weight loss at an oral dose of 1mg/kg/ day. At higher doses, LGD-3303 significantly increased muscle weight ahead of eugonadal levels. However, potency and efficacy at other measurement endpoints were greatly reduced. LGD-3303 does not maintain eugonadal levels of serum LH at doses below 10mg/kg/ day, maintains prefrontal frontal weight only at doses of 100mg/kg/ day or higher, and does not return average prepuce gland weight to eugonadal levels at any of the tested doses
