    Publisher SummaryDibucaine is 2-butoxy-N-[2-(diethylamino)­ethyl]-4-quinolinecarboxamide. This chapter discusses the physical and chemical properties, synthesis, stability-degradation, drug metabolism and pharmacokinetics, toxicity, and methods of the analysis of dibucaine. The proton nuclear magnetic resonance (NMR) spectra of dibucaine and dibucaine hydrochloride are presented in the chapter. The spectra have been determined on a Perkin–Elmer R-24B 60 MHz spectrometer at ambient temperature. The samples were dissolved in deuterated chloroform containing tetramethylsilane as an internal standard. Differential scanning calorimetry (DSC) thermogram of dibucaine shows one endotherm between 61° and 70°C with a melting point of 64°C and a purity value of 98.9 mole percent, when the thermogram is followed in a DuPont Model 900 instrument at a scan rate of 20°C/minute. Similarly, the DSC thermogram of dibucaine hydrochloride shows one endotherm between 94° and 104°C with a melting point of 97°C. The purity of the hydrochloride has been determined to be 99.4 mole%. Two identification tests are given in the USP XX for dibucaine—one an infrared absorption test and the other an ultraviolet absorption test.
