SummaryWe describe a double blind trial of lymecycline, demethylchlortetracycline and methacyline in 190 patients suffering exacerbations of chronic bronchitis with purulent sputum. The dose given of all three compounds was 1200 mg or its equivalent daily for fourteen days. The three compounds were equal in clinical effect. Unwanted effects appeared in nearly half of those treated with demethylchlortetracycline, in over a third given methacycline and in less than a sixth given lymecycline, sufficient to cause changes in treatment in most affected.Taking account of these results and a previous comparison of lymecycline and tetracycline, and considering the relative costs of the various compounds, we suggest that in purulent exacerbations of chronic bronchitis tetracycline hydrochloride or oxytetracycline should be used first if a high dose of tetracycline is required and replaced by lymecycline if intolerance appears.
