The effect of a new thyroid component, recently isolated from bovine thyroglobulin. on the respiration and swelling of rat liver mitochondria, has been studied. This product is an iodinated peptide, 3,5 diiodo-l-tyrosyl 3,5 diiodo-l-tyrosine (TT4I) and its action is quite different from that of iodothyronines which act as uncouplers and inhibitors of mitochondrial respiration. 3,5 diiodo-l-tyrosyl 3,5 diiodo-l-tyrosine is an inhibitor of mitochondrial respiration. It acts on the three coupling sites of oxydo-phosphorylations with various intensities. Furthermore, it is able to prevent mitochondrial swelling induced by thyroxine. It seems that the hypothesis that this compound has thryoximimetic properties is incorrect since its peripheral effects are quite different from those of thyroid hormones.
