High pressure liquid chromatography and gas chromatography-mass spectrometry revealed that hamster liver microsomes form N-hydroxyacetaminophen from N-hydroxyphenacetin (cas 19315-64-1) but apparently not from acetaminophen. Nevertheless more covalent binding of radiolabel occured with 3H-acetaminophen as a substrate than with 14C N-hydroxyphenacetin. It, therefore, seems likely that the principle chemically reactive metabolite formed from acetaminophen does not arise through the formation of N-hydroxyacetaminophen as has been previously postulated.
