1. A stacking interaction of 6-methoxypurine in water has been demonstrated by analysis of NMR chemical shift data. The compound is an example of a substituted purine whose solubility in water is too low to yield significant data by itself. However, a technique is described for studying the compound in the presence of a solubilizing agent, in this instance, purine.2. The solubility of 6-methoxypurine can be increased from 0.009 M in water to over 0.1 M in the presence of excess purine.3. Increasing purine concentration in low ranges, or decreasing temperature, produce an upfield chemical shift for 6-methoxypurine. Both of these are interpreted as increasing a stacking interaction. The upfield shifts are not observed in nonaqueous solvents.
