One of the reported advantages of the avicide 2-chloro-4-acetotoluidide (CAT) over its parent compound, 3-chloro-p-toluidine (CPT, DRC-1339) is its markedly lower oral toxicity in mammals. The present study was undertaken in an effort to elucidate the reasons for the differences in toxicity of these two compounds and to determine the effect, if any, of the solvent previously used (dimethyl sulfoxide) on the toxicity of CAT. To these ends, the lethal doses of CPT and CAT, in different vehicles and via various routes of administration, were determined and compared. The results indicate that the relatively low oral toxicity of CAT may be due to poor absorption by the gastrointestinal tract. The lack of cyanosis in the rats used for this study gives additional support to the previous observation that CAT is devoid of methemoglobin-inducing properties in this species (and apparently in rabbits).
