betazole (cas 105-20-4), (β-(3-pyrazolyl)-ethylammonium), a very weak H2-receptor agonist of histamine is compared, in the semiempirical quantum chemical CNDO/2 approximation, to histamine.The molecular determinants responsible for the H2-activity, such as the conformation of the ethylammonium side chain, a negative global charge on the imidazole ring, the existence of a proton on the N-terminal, as well as the inversion of the tautomeric preference for the cationic and neutral forms are considered. It was determined that betazole fulfilled all of the above except for the tautomerism N3-H→N1-H. It is concluded that the proton transfer from N3 to N1 is an important and necessary step in the mechanism of action of the H2-receptor agonists of histamine.
