Edeine (cas 11006-90-9) inhibits the synthesis of bacterial DNA as shown by the incorporation studies of [3H]thymidine into DNA of Escherichia coli 15 T−. It does not induce a release of [3H]thymidine from prelabeled DNA. Edeine does not bind to the DNA. DNA isolated from edeine treated Bacillus subtilis shows the same binding pattern as the DNA of the untreated control, when centrifuged to the equilibrium in a CsCl density gradient. The resulting DNA heat denatured and rapidly cooled bands in the same position as the denatured normal DNA. The specific transforming activity of DNA obtained from B. subtilis exposed to the growth inhibitory concentration of edeine is the same as the transforming activity of the control DNA. Prolonged action of edeine added in large amount to the bacterial cultures decreases it. Edeine does not form complexes in vitro with poly-U and poly-A.
