Publisher SummaryThis chapter focuses on the CHROMIUM-51 (cas 14392-02-0) radioimmunoassay for chemotaxis. Subsequently, it has been modified for use with monocytes and other leukocytes, and some laboratories have used microassay to minimize the amount of material employed. In this assay, leukocytes are labeled with chromium-51 and then placed in the upper compartment of a chemotactic chamber, which is separated from a lower compartment by two filters. A major limitation of the radioassay of chemotaxis is that relatively large numbers of cells are used. This renders the technique impractical for the study of neutrophils from neonates and small children. However, recently the radioassay method has been modified for use with multiwell microchamber. Although the microassay is appealing and may be useful in settings of limited chemoattractant or cells, it has not yet been subjected to sufficient evaluation to know its reliability. The radioassay of leukocyte locomotion has proved a reliable functional screening test for clinical studies, which for some diseases is more reliable than many of the other available assays. The method is easy to learn and eliminates the time-consuming task of the microscopic enumeration of leukocytes in stained filters.
