The structure of the monosaccharide isolated from mycoside G, a specific glycolipid of Mycobacterium marinum, is established as follows: demethylation experiments show that the monosaccharide is a methyl ether of l-rhamnose. Spectra (mass and n.m.r.) of the derived, acetylated methyl glycoside suggest a 6-deoxy-3-O-methylpyranoside structure, which is confirmed by periodate oxidation. The l-configuration of the sugar is deduced from the optical rotation, which is identical with that of 3-O-methyl-l-rhamnose (acofriose). The circular dichroism curve (negative Cotton effects) of the acetylated mycoside establishes the existence of an α-l-glycosidic link between the sugar and the aglycon. Mycoside G is therefore a 3-O-methyl-α-l-rhamnopyranoside of a p-substituted phenol.
