    To confirm the absence of trans fatty acids, quantitative methods are required for the determination of cis and trans fatty acids. Simultaneous quantification of oleic and elaidic acids, which are cis–trans isomers, in mixed solutions was realized using Raman spectroscopy. The Raman spectra of oleic acid and elaidic acid are very similar, except for a band in the 1600 cm−1 region that is assigned to the stretching vibration mode of CC bonds. Partial least squares (PLS) regression was used for the determination of these fatty acids, and the predicted and actual values were nearly identical to the actual values. To confirm the reproducibility of this method, the Raman spectra of mixed solutions with known concentrations were recorded. The concentrations of oleic and elaidic acids predicted by the PLS model were in good agreement with the actual concentrations. Thus, Raman spectroscopy with multivariate analysis is a useful method for the simultaneous determination of cis–trans fatty acids.
