Protein misfolding, aggregation and amyloid fibril formation lead to a number of human diseases. Monitoring the fibrillation of proteins and interfering the formation of the disease associated protein fibers are among the most important tasks in the field. Herein, we describe that bromophenol blue (BPB), a well-known conventional indicator for electrophoresis, can serve as a novel probe for detection and inhibition of amyloid proteins. In this study, insulin was used as a model amyloid protein, while BPB served as a protein isomerization targeting (PIT) dye that emits fluorescence upon binding to protein fibers. By adding trace amount of BPB, the three stages of the insulin fiber formation can be revealed. When higher concentration of BPB was present, the fibrillation process was largely blocked, indicating BPB potential role as an inhibitor of protein amyloidosis. This study opens an avenue for the development of new probes and inhibitors of amyloid proteins using PIT dyes.
