Recently, the cis-isomer of zeatin riboside (ZR) was reported as a free cytokinin in tobacco shoots as well as in a few other plant extracts. Although the identification was based on physico-chemical methods, the possibility that it was an artefact of extraction procedure used was not excluded. In this report the extraction procedure used in the above studies was compared with a procedure known to minimise substantially any extraction arteacts. Using deuterium labelled internal standards and gas chromatography-mass spectrometry in the multiple ion detection mode (GC-MS-MID) the levels of ZR and cis-ZR were quantified in tobacco shoot extracts. Although the levels of ZR determined by the two extraction procedures were in very close agreement, widely differing results were obtained for the cis-ZR. In contrast to earlier reports, we conclude that cis-ZR does not occur as a free cytokinin in tobacco shoots.
