  • Estimation of sectioning compression and thickness of ultrathin sections through Vestopal-W-embedded cat spinal roots
  • Add time:07/13/2019         Source:sciencedirect.com

The degree of sectioning compression and the thickness of ultrathin sections were studied in Vestopal-W-embedded feline spinal roots using a conventional sectioning technique. Sections about 100 nm thick were found to have been compressed by ∼5% in the direction of sectioning. No corresponding widening was detected in the perpendicular direction. There were no significant signs of a differentiated compression or widening within single sections. Mean section thickness was obtained from the height of a trimmed-out specimen mesa and the number of sections cut from it. Using this mean value and the mean section electron-scattering value, as estimated in the electron microscope, it was possible, in a single section about 100 nm thick, to calculate the relative thickness with an uncertainty of less than 10%.
