Publisher SummaryThis chapter describes the preparation and purification of PTEROIC ACID (cas 119-24-4) from pteroylglutamic acid (folic acid). Pteroic acid can be prepared conveniently, by the removal of glutamic acid, from commercially available pteroyiglutamic acid, using a pteroylglutamic acid-degrading pseudomonad. In this study, for the preparation of crude pteroic acid, the pteroylglutamic acid-fermenting pseudomonad (ATCC 25301) is inoculated into glass universal bottles, containing 10 ml of sterilized medium, made by dissolving 5 g KH2PO4 and 12 g K2HPO4·3 H2O in 900 ml distilled water and adding to it 5 ml and 1.5 ml of two stock mineral salt solutions that are: (l) 1% MgSO4, 0.1% CaCl2·H2O and 0.1% FeSO4 7 H2O, the supernatant after two days being used, and (2) 0.1% MnSO4 and 0.1% Na2MbO4 2 H2O, the solution being used directly. To the above is added l0 g of pteroylglutamate dissolved prior to the addition in 0.01 N NaOH and readjusted to neutrality prior to the addition. The solid pteroic acid obtained, upon analysis in a paper chromatography system, shows a complete absence of impurities. Similarly, when analyzed on TLC, a complete absence of fluorescent impurities is found. Its authenticity has been established by ultraviolet (UV) and infrared spectra and by microbiological assay.
