Poly 4-methyl- and poly 4,4-dimethyl-cytidylic acids have been prepared by the action of polynucleotide phosphorylase (from Micrococcus lysodeikticus) on the corresponding nucleoside 5′-pyrophosphates. Random co-polymers containing cytosine and 4-methyl- or 4,4-diamethyl-cytosine residues have also been prepared.Poly 4-MeC† and poly 4,4-diMeC were both completely hydrolysed in the presence of pancreatic ribonuclease and venom phosphodiesterase to the corresponding 3′- and 5′-nucleotides respectively. The secondary structures of these two polymers differ appreciably from that of poly C under acidic, but not under neutral, conditions. Poly [C, 4-MeC (39%)] forms a 1:1 complex with poly I. Both poly[C, 4,4-diMeC (13%)] and poly[C, 3-MeC (15%)] form low-melting complexes with less than one part of poly I.
