Hamycin, either alone or in suspension as hamycin-cholesterol complex, was intravenously administered to mice in a single dose. Plasma antifungal activity at different time intervals and LD50 at varying drug: cholesterol proportions were studied. Hamycin alone was eight-fold more toxic than the hamycin-cholesterol complex. While the antifungal activity of hamycin in plasma rapidly declined by first order kinetics, the activity of hamycin-cholesterol complex in plasma was maintained at the initial high level for a prolonged period and with a 12-fold increase in the apparent volume of distribution of the drug. The therapeutic implications of these findings are discussed.
