Publisher SummaryThis chapter focuses on the synthesis of D-Galactose 6-phosphate and D-tagatose 6-phosphate. D-Galactose 6-phosphate and D-tagatose 6-phosphate are intermediates in the catabolism of lactose and D-galactose in Staphylococcus aureus and in group N streptococci. D-Tagatose 6-phosphate is also an intermediate in galactitol catabolism in Klebsiella pneumoniae and in Escherichia coli. D-Galactose 6-phosphate is used as a substrate in the assay of D-galactose-6-phosphate isomerase, whereas D-tagatose 6-phosphate is used as a substrate in the assay of D-tagatose-6-phosphate kinase, in the reverse reaction of D-galactose-6-phosphate isomerase, and as a substrate-precursor in the assay of D-tagatose-l,6-bisphosphate aldolase. D-Galactose is multiply phosphorylated with polyphosphoric acid. The most resistant phosphate groups are then removed by acid hydrolysis, leaving primarily D-galactose 6-phosphate, but also other positional isomers of D-galactose monophosphate. D-Galactose 6-phosphate is purified, free of the positional isomers, by ion-exchange chromatography. The synthesis of D-tagatose 6-phosphate from 1,2:3,4-di-O-isopropylidene-D-tagatose involves the isomerization of D-galacturonic acid to D-tagaturonic acid, or the preparation of 1,2 : 3,4- di-O-isopropylidene-D-tagaturonic acid.
