Publisher SummaryLysyl oxidase activity and the rate of cross-linking must be reduced effectively to cause an increase in the net pool of soluble elastins prior to isolation. This may be achieved by rendering animals nutritionally copper-deficient or lathrytic. The choice of experimental animals is also important. In this regard the chick system has been of fundamental importance because the isolation of soluble elastin from other common experimental animals, such as rats and mice, has proved to be difficult. If a nutritional or pharmacological treatment is used to precondition animals for subsequent isolation of soluble elastin, it is often necessary to initiate the treatment during periods of neonatal development. The chick is easily manipulated nutritionally during neonatal development, and with a sufficient number of chicks the yield of thoracic aorta, an excellent source of soluble elastin, is adequate for soluble elastin isolation.
