    Publisher SummaryThis chapter describes the physical properties and methods of analysis of methadone hydrochloride. The absolute configuration of the molecule is determined by measuring the effect on two selected sets of reflexions of the imaginary part of the dispersion of copper radiation by the bromine atom. The methods of analysis of the molecule include gravimetric analysis, ultraviolet (UV) analysis, ion-exchange chromatography, radio-immunoassay, thin-layer chromatography (TLC), gas chromatography, and high-pressure liquid chromatography (HPLC). The UV analysis is based on the oxidation of the drug with alkaline potassium permanganate to form benzophenone. Gas chromatography involves the extraction of a tablet at 37° with successive portions of dissolution medium. The high sensitivity and the low quantities of drug detected by HPLC method indicates that this method may be successfully used for the in vivo determination of methadone.
